Bacteria room

The bacteria room provides all the instruments needed for the culture, genetic engineering, selection and expansion of bacteria.

Before using the instruments for the first time, contact the trainers below for mandatory training.
Users are expected to perform experiments independently and must be adequately trained to ensure proper use of the instruments.

How to make reservation

Prior to use, it is mandatory to reserve instruments through the booking platform of the Department of Biomedical Sciences.

Before your session
  • Wear a lab coat and clean gloves.
  • Equipment must be checked in all their parts: everything must be clean and without any obvious damage. If this is not the case, please immediately contact the staff.
  • CHECK IF YOU ARE THE FIRST USER OF THE DAY AND, IN THIS CASE, TURN ON THE UV LAMP FOR AT LEAST 20 MINUTES. REMEMBER TO TURN OFF THE UV LAMP BEFORE USING THE HOOD!
  • MANDATORY HOOD OPENING PROCEDURE: switch on the hood and the flux; ONLY once the flux is fully operational, the frontal lid can be removed.
  • Spectrophotometer must be switched on 10 minutes before use.
  • Report to the staff the presence of incorrect behavior or waste not properly disposed of.
During your session

Good lab practices must be followed. Perform procedures in a way that minimizes aerosols (opening tubes slowly, pouring liquids without splashing, etc.).

The agar plates have to be labeled with a NAME and can be incubated inside the indicated drawers, in the room at 37°C.

After your session
  • Clean all surfaces first with water and then with 70% ethanol. Please do not use bleach on any work surface.
  • Properly discard solid biological waste into a biological bin of your lab. It can be left inside the room, LABELED WITH A NAME AND OPENING DATE and CLOSE OUT OF YOUR SESSION. Don’t leave open bins for long periods. The collection and conferral to the office of competence remains the RESPONSIBILITY OF THE PRODUCER.
  • All liquid biological waste must be bleached (1:10) and DISCARDED IN YOUR OWN LAB.
  • DO NOT leave anything in the room (out of your assigned space in the shelf) and inside the hoods and thermoshakers.
  • Switch off the spectrophotometer after each use, to prevent deterioration of the light source.
  • Switch off thermoshakers and check if the surfaces and positions are clean. If not, it must be cleaned first with water and then with 70% ethanol or denatured alcohol.
  • Switch off the centrifuge after each use.
  • MANDATORY HOOD CLOSING PROCEDURE: the frontal lid must be positioned back in place BEFORE switching off the flux; ONLY once the hood is properly closed, switch off the flux. Switch off the light and THEN the hood.
  • Contaminated gloves must be discarded.

Specific indication for this facility

In the booking webpage you can find hoods and a separated section for shakers.

Please note that the shaker New Brunswick Scientific C24 can be used only for temperature ≥30°C.

Shakers Orbi-shaker Benchmark and Ika Labortechnik KS 125 BASIC are located in the room at 37°C (room 69/GEOTEC 06-128).

For shakers Excella E25, Innova 44, New Brunswick Scientific C24, Orbi-shaker Benchmark and Ika Labortechnik KS 125 BASIC bottle positions are identified with a code and need to be BOOKED INDIVIDUALLY.

The letter of the code indicates the size of the position (XS/S/M/L/XL). You can find the picture of the positions of the shakers in the pdf document on the webpage and attached in the Bacteria Room. Please use the code of the booked positions.

As different cultures could need different temperatures of incubation, in case of more reservations at the same time THE TEMPERATURE OF THE SHAKER IS DEFINED BY THE FIRST USER BOOKED, that will specify the protocol in use in the description field. Each bottle has to be labeled with NAME, LAB, TELEPHONE NUMBER of the owner.

The same user can’t book multiple shakers at the same time, to allow everyone to use them.
